On the local (Columbia, SC) radio station they were discussing this and apparently former Gamecock Chris Culliver (yeah, the one with the gay remark at the Super Bowl) said one team asked him if he had ever worn women's panties.
Definitely some strange questions there, but they're usually probably more interested in your reaction than what you actually say.
The whole thing is beginning to get a little silly. I had to start questioning my own sexuality after I realized I was watching hulking, cut dudes in neon spandex basically exercising for hours on end.

Great. Now I know what puce is after reading this post. Apparently I'm going to fail (or pass depending on your point of view) the gay test from here on out. My wife is going to be pissed.

What? Football players have contracts but that doesn't make them "contract workers" in the legal sense. They're employees of the team with which they sign a contract for every purpose I know of (especially federal tax laws, which is one of the most significant areas where the difference between "employee" and "independent contractor" is important).
As for asking the question about being gay, in the context of the Combine I think it depends on State law and how strict a given State's employment/discrimination laws are. I read somewhere that 17 States have laws stringent enough to prevent this question from being asked and that it would be o.k. in the others.
I'm not an employment/labor lawyer so I don't know any more about the second paragraph of this message than anyone else. If I'm wrong feel free to correct me, my feelings won't be hurt.
